A 2018 study broke neuroticism apart into its 12 individual questionnaire items and studied each one's genetics separately — "irritable mood" is one of them, and it turns out not to be genetically identical to the rest of neuroticism.
Most genetic studies of personality and mood measure a single composite score — for neuroticism, that usually means summing answers across a whole questionnaire into one number. Nagel et al. 2018 asked a different question: what if the individual items that make up that questionnaire aren't all measuring the same underlying genetics?
The study ran genome-wide association analyses on each of 12 individual neuroticism questionnaire items — including irritable mood — separately, across two UK Biobank samples (roughly 106,218 to 109,017 people in the first, 260,083 to 266,896 in the second), then combined the results in a meta-analysis. Genetic correlations between the 12 items ranged from 0.38 to 0.91 (average 0.63) — related to each other, as expected, but far from identical. The combined analysis found 255 genome-wide-significant independent genomic regions in total, and 138 of them were specific to just one item rather than shared broadly across all twelve. Clustering the items by their genetic correlations revealed two more genetically distinct groups within neuroticism, which the authors labelled "depressed affect" and "worry."

The practical conclusion the authors draw is that "neuroticism," as usually measured and studied, is not one genetically uniform thing — it's a bundle of related but distinct items, and some of the biology only becomes visible when the items are examined one at a time instead of averaged together into a single score.
